Nuclear Binding of Progesterone in Chick Oviduct
نویسنده
چکیده
1. Varied doses of labelled or unlabelled progesterone were injected into immature chicks which had previously been stimulated with oestrogen. The concentrations of nuclear bound [3H]progesterone were correlated with the effects of the hormone on endogenous RNA polymerase I and II activities in isolated oviduct nuclei. 2. The extent of nuclear localization of [H]progesterone in oviduct (a progesterone target tissue) was shown to be much greater than in lung (non-target tissue). The concentration of bivalent cations in solvents used in the nuclei isolations has a marked effect on the amount of bound hormone in the nuclei. 3. Evidence for the existence of several classes of binding sites for progesterone in the oviduct nuclei is given. These classes represent about 1000, 10000 and 100000 molecules of the hormone per cell nucleus and are saturated by injecting approx. 10, 100 and 1000g of progesterone respectively. 4. When saturation of the first (highest affinity) class ofnuclear sites occurs, a marked inhibition in RNA polymerase II (but not RNA polymerase I) activity was observed. When the second class of sites was saturated, alterations in both RNA polymerase I and II activities were observed. Binding to the third class of nuclear binding sites was not accompanied by further changes in polymerase activity. It is suggested that the first two classes of nuclear binding sites may represent functional sites for progesterone action in the chick oviduct.

Progesterone "receptors" in the cytoplasm and nucleus of chick oviduct target tissue.
This report demonstrates that the chick oviduct, a specific target organ for progesterone, contains both cytoplasmic and nuclear macromolecules which bind progestins. These binding molecules can be clearly distinguished from transcortin by centrifugation through sucrose gradients of low ionic strength and by agarose gel filtration. The cytoplasmic progesterone-binding molecules also bind 5-alph...

The cytoplasmic progesterone-binding proteins of chick oviduct are shown to consist of two major 4 S fractions which occur in equal amounts. They are separated from each other by DEAE-cellulose chromatography. Purification of the two components, A and B, was about 800and 3000-fold, respectively, from estrogen-primed chick oviducts following ultracentrifugation, ammonium sulfate precipitation, g...

Glucocorticoids induce ovalbumin and conalbumin mRNA in oviducts from chicks withdrawn from prior estrogen treatment. The magnitude and the kinetics of the responses obtained, either in vivo or in vitro, are comparable to those obtained with estrogen or progesterone. With cultured oviducts, 1 nM dexamethasone is sufficient for half-maximal accumulation of nuclear receptors and partial induction...

To study the promoter of the chicken progesterone receptor (cPR) gene and the relevance of several progestin-responsive elements therein, chimeric genes were constructed which contained the 5'-flanking region of the cPR gene linked to promoterless globin or chloramphenicol acetyltransferase sequences. Cell-specific initiation of transcription was observed in transiently transfected chicken embr...

RNA was extracted from oviduct, shell gland, and liver of chicks treated with diethylstilbestrol and a combination of diethylstilbestrol and progesterone. Preparations containing 500 or 2500 mug of RNA were put in the oviductal lumen of diethylstilbestrol-treated chicks for 24 hr, and the avidin content of the oviduct was determined by measurement of [(14)C]biotin binding. The results of the pr...
